No, the concept of using a mod manager is to basically make your life easier, you just need one and you must only use 1 either Mod organizer or NMM, both are fine to use, nmm isnt exclusive for mods at nexus (though its download platform is AWESOME, I mean the speed of download as being linked to the nexus page is far superior than M.O) BUT NMM overwrites directly on the data folder of skyrim
